    LH Beta in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of LH Beta protein in mouse tissue extract samples.

    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    LH Beta is a polypeptide synthesized by gonadotropic cells. Together with the alpha subunit, they form the heterodimeric luteinizing hormone, which stimulates ovulation and testosterone synthesis. Urinary luteinizing hormone levels are used to predict fertility timing. Dysregulation of this system leads to premature menopause, hypogonadism, and is associated with a host of sexual development disorders.

Ab235648 mouse LH beta Elisa kit was tried for LH in media of tissue culture and in mouse serum.
Samples included in trial:
5ul from standard stock (192ng/ml) were added to 110ul of cell culture media.
The same media was used to verify background.
Media from primary gonadotrophs cells was taken 20 minutes after 1 minute treatment with 1nM GnRH (or mock).
Serum was taken from 47 days old female mice at estrus.
All samples included 1x cell extraction enhancer solution. (2.2ul of x50 was added to 110ul sample).
All samples and standards were measured as 50ul duplicates.
Incubation with the antibody mix was for 2 hr at RT.
The rest of the protocol – as in the manual.
Calculation – every well was read 3 times and averaged.
every duplicate was averaged.
standard tube #8 (no protein added) served as blank and was subtracted from each result.
standards up to 938pg/ml were linear and their equation was used for LH concentration calculation.
values above that – were estimated by proximity to the relevant standard.
Under these conditions, LH could be detected in both serum and media.
