  • Product overview

Abcam’s MCP1 (CCL2) in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of MCP1 protein in mouse serum, plasma and cell culture supernatants

The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

  • Notes

MCP1 (CCL2) is a chemokine expressed by fibroblasts, macrophages, endothelial cells, mast cells, osteoblasts, ameloblasts, epithelial cells, and astrocytes after stimulation with inflammatory stimuli. MCP1 is involved in the recruitment of monocytes and lymphocytes and in the regulation of Th1/Th2 lymphocyte differentiation.

  • Research areas

  • Function

Chemotactic factor that attracts monocytes and basophils but not neutrophils or eosinophils. Augments monocyte anti-tumor activity. Has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis or atherosclerosis. May be involved in the recruitment of monocytes into the arterial wall during the disease process of atherosclerosis.
  • Sequence similarities

Belongs to the intercrine beta (chemokine CC) family.
  • Post-translational

Processing at the N-terminus can regulate receptor and target cell selectivity. Deletion of the N-terminal residue converts it from an activator of basophil to an eosinophil chemoattractant.

Application Abreviews Notes
Sandwich ELISA Use at an assay dependent concentration.

  • SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

  • Standard curve comparison between mouse MCP1 SimpleStep ELISA® kit and traditional ELISA kit from leading competitor. SimpleStep ELISA kit shows a 3-fold increase in sensitivity.

  • Background-subtracted data values (mean +/- SD) are graphed.

  • The concentrations of MCP1 were measured in duplicates, interpolated from the MCP1 standard curves and corrected for sample dilution. 1X diluted samples are as follows: serum 17.5%, plasma (citrate) 40%, plasma (heparin) 40% and plasma (EDTA) 40%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MCP1 concentration was determined to be 245.9 pg/mL in serum, 159.7 pg/mL in plasma (citrate), 215.7 pg/mL in plasma (heparin) and 193.9 pg/mL in plasma (EDTA).

Abreview for ab208979 - Mouse MCP1 ELISA Kit

Excellent Excellent 5/5 (Ease of Use)
Assay was performed using the Abcam Kit protocol. Blood samples were obtained from 15-17 week-old male C57Bl/6 mice and serum samples were prepared and stored as recommended in the Abcam Kit protocol. Serum samples were analyzed diluted 1:32 times. All sample concentrations were within the kit detection range. CCL2 serum level in healthy control group was 115±77 pg/mL.

Figure 1: Box-and-whisker plot of MCP1 serum levels in control (n=5) and collagen-induced arthritis (CIA) group (n=6) 35 days after primary immunization with chicken type II collagen.
