ELISA
To control the general presence of IgG isotypes (IgG1, IgG2, IgG3, IgG4) and to check for IgEs and IgAs within purified total-IgG samples from serum ELISAs were performed as follows. Purified IgG fractions from 8 ERT-naive patient samples (patients 1 to 8) were pooled and 5.0 µg IgGs were immobilized on 96-well Maxisorp ELISA plates (Thermo Fisher Scientific) overnight at 48C, washed 3 times with PBS/Tween 0.1% and blocked with 100 µl PBS/BSA 2% for 2 hours at RT. Subsequently, 0.25 mg of IgG isotype-specific secondary antibodies (mouse a-human IgG isotype 1[ab99774] mouse a-human IgG isotype 2 [ab99779] rabbit a-human IgG isotype 3 [ab86253] mouse a-human IgG isotype 4 [ab99823] goat a-human IgA [ab97215] mouse a-human IgE [ab99806] Abcam, Cambridge, UK) were applied to the samples (in 100 µl PBS) and incubated for an additional 2 hours at RT. Rabbit a-human IgG isotype 3 was detected with goat a-rabbit IgG-HRP [sc-2054, Santa Cruz Biotechnology] as secondary antibody (diluted 1:5000). After 5 additional washes, 100 µl 1-Step Ultra TMB-ELISA Substrate Solution (Thermo Fisher Scientific) was added to each well, incubated for 10 minutes and stopped with 100 µl 2 mol/L sulfuric acid. Absorption was measured at 450 nm and calculated per microgram total IgG. Results demonstrated the measurable presence of IgG isotypes 1 to 4 within purified IgGs as well a moderate presence of IgAs of ERT-naive patients.
