    PD-1H in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of PD-1H protein in mouse serum, plasma, and cell and tissue extracts.

    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    Programmed Death-1 Homolog or PD-1H, also known as V-type immunoglobulin domain-containing suppressor of T-cell activation (VISTA), B7-H5, and Dies1, is a member of the immunoglobulin superfamily proteins and is encoded by the gene Vsir. PD-1H is a 308- amino acid type 1 glycosylated transmembrane protein that is primarily expressed in white blood cells and the transcription is partially controlled by p53. PD-1H has been shown to act as ligand and a receptor on T-cells to inhibit effector function. Increased PD-1H expression is related with an increase in immune activation and a decrease in CD4-posititive T-cells.

  • The PD-1H standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

  • The concentrations of PD-1H were measured in duplicates, interpolated from the PD-1H standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%, plasma (EDTA) 100%, and plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-1H concentration in the neat samples was determined to be 19.7 ng/mL in serum, 17.8 ng/mL in plasma (citrate), 15.3 ng/mL in plasma (EDTA), and 31.8 ng/mL in plasma (heparin).

  • The concentrations of PD-1H were measured in duplicate and interpolated from the PD-1H standard curve and corrected for sample dilution. Undiluted samples are 250 µg/mL for all extracts. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
