    Abcam’s RANKL Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse RANKL in serum, plasma and cell culture supernatants.

    This assay employs an antibody specific for Mouse RANKL coated on a 96- well plate. Standards and samples are pipetted into the wells and RANKL present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse RANKL antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of RANKL bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

  • Function

    Cytokine that binds to TNFRSF11B/OPG and to TNFRSF11A/RANK. Osteoclast differentiation and activation factor. Augments the ability of dendritic cells to stimulate naive T-cell proliferation. May be an important regulator of interactions between T-cells and dendritic cells and may play a role in the regulation of the T-cell-dependent immune response. May also play an important role in enhanced bone-resorption in humoral hypercalcemia of malignancy.
  • Tissue specificity

    Highest in the peripheral lymph nodes, weak in spleen, peripheral blood Leukocytes, bone marrow, heart, placenta, skeletal muscle, stomach and thyroid.
  • Involvement in disease

    Defects in TNFSF11 are the cause of osteopetrosis autosomal recessive type 2 (OPTB2) [MIM:259710]; also known as osteoclast-poor osteopetrosis. Osteopetrosis is a rare genetic disease characterized by abnormally dense bone, due to defective resorption of immature bone. The disorder occurs in two forms: a severe autosomal recessive form occurring in utero, infancy, or childhood, and a benign autosomal dominant form occurring in adolescence or adulthood. Autosomal recessive osteopetrosis is usually associated with normal or elevated amount of non-functional osteoclasts. OPTB2 is characterized by paucity of osteoclasts, suggesting a molecular defect in osteoclast development.

We ran the RankL Elisa kits with media from cells that had been irradiated or non irradiated and a treatment group aa. Radiation should cause an increase in RankL which it did was seen in the Elisa.

We followed the instructions in the kits and corrected the plotted data with BCA concentrations.

Mouse RANKL ELISA for OA

RANKL evaluation was performed on Plasma isolated from blood samples to determine OA onset.
Our Samples included: Groups with joint damage, groups with joint damage and walking, and a normal group.
Assay procedure was performed according to manufacturer instructions. We had to dilute our samples according to our BCA results to what the manufacturer suggested. The Elisa kit is very straight forward, with the normal steps. Readings were performed at 450 nm. Photos show the results of standard curve and Test run for the kit.
This kit very simple to use if you have a basic knowledge of an Elisa. Our lab will be using more of these

This product has to our knowledge not been tested on human serum samples. However the sequence homology between the two species is such that it is possible the kit may work. Question

A while ago we tested the RANKL ELISA kit (ab100749). Our assay did not work. There was no quantifiable levels of RANK in our samples.

We would like to come back to this experiment and therefore would like to troubleshoot with you. We used a 10X sample dilution. Our sample readings were very low, though our standard wells were perfect.

First off, we use mouse plasma, collected in EDTA treated tubes. The kit insert does not mention whether certain anti-coagulant interfere. Can we assume EDTA does not counteract?

What are expected values for RANKL using this kit?

I have confirmed with the laboratory that EDTA will not interfere with this kit.When we tested normal mouse serum and plasma samples with this kit, the measured levels were around 300 pg/ml. You will have to test several different dilutions, as the amount of RANKL can vary from sample to sample. I would recommend a range of neat through 1:5.
