    Abcam’s SCF Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse SCF in serum, plasma and cell culture supernatants.

    This assay employs an antibody specific for mouse SCF coated on a 96-well plate. Standards and samples are pipetted into the wells and SCF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse SCF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of SCF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

  • Function

    Ligand for the receptor-type protein-tyrosine kinase KIT. Plays an essential role in the regulation of cell survival and proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and in melanogenesis. KITLG/SCF binding can activate several signaling pathways. Promotes phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase, and subsequent activation of the kinase AKT1. KITLG/SCF and KIT also transmit signals via GRB2 and activation of RAS, RAF1 and the MAP kinases MAPK1/ERK2 and/or MAPK3/ERK1. KITLG/SCF and KIT promote activation of STAT family members STAT1, STAT3 and STAT5. KITLG/SCF and KIT promote activation of PLCG1, leading to the production of the cellular signaling molecules diacylglycerol and inositol 1,4,5-trisphosphate. KITLG/SCF acts synergistically with other cytokines, probably interleukins.

LMW-SCF is fully N-glycosylated at Asn-145, partially N-glycosylated at Asn-90, O-glycosylated at Ser-167, Thr-168 and Thr-180, and not glycosylated at Asn-97 or Asn-118. HMW-SCF is N-glycosylated at Asn-118, Asn-90 and Asn-145, O-glycosylated at Ser-167, Thr-168 and Thr-180, and not glycosylated at Asn-97.
    A soluble form exists as a cleavage product of the extracellular domain.

      Thank you for your inquiry.

      Unfortunately, we do not have any cross reactivity data for testing rat samples using the mouse stem cell factor ELISA kit (ab100740). Based on online literature though, mouse SCF has ˜93% homology with rat SCF so there is certainly a good chance of getting cross reactivity. However, the only way to know for sure would be to test. If it helps, both the capture and detection antibody in ab100740 are polyclonal goat IgG. While ab100740 is neither designed nor validated with tissue lysate samples, our kits are very accommodating to many different samples types so lysates will most likely work. We recommend diluting the samples at least 5-fold with Assay Diluent B (Item E) to minimize any effects of the detergents in the lysis buffer. The samples may need to be diluted further but this would need to be determined by the the end-use empirically.

      Basically, a lysis buffer must meet the following specifications:

      A) has relatively low salt content (700 mM or less)
      B) does not contain sodium azide
      C) does not contain more than 0.1% SDS
      D) does not contain more than 10 mM reducing agents (beta-mercaptoethanol or dithiothreitol).

      This would include any buffers used for immunoprecipitations, including RIPA buffer.

      First, I want to apologize to you for the delay in gathering this information. I hope this did not cause you any inconveniences.

      In regards to using rat tissues in our SCF Mouse ELISA kit, there is no cross reactivity data testing this mouse kit (ab100740) with rat samples so we do not know. However, we do know that the antibody pair is binding somewhere on the extracellular domain of the SCF protein and that mouse and rat share ˜93% homology on the extracellular domain. Therefore there is a good chance that there will be cross reactivity. Nevertheless, the only way to be certain would be to test.



      As to your questions about using urine with ab100786 and ab119602.

      Ab119602 will work with urine future datasheets will include this.

      While ab100786 was not validated with urine samples (only serum, plasma, and cell culture supernatants) these types of kit are very accommodating to many types of samples so I see no reason why urine or any other body fluid for that matter would not be compatible with this kit.
