Question
Answer

Thank you and your customer for taking the time to complete our questionnaire and contact us. I am sorry to hear your customer has had difficulty obtaining satisfactory results from this antibody.

The details your customer has kindly provided will enable us to investigate this case for your customer and this is also helpful in our records for monitoring of quality.

Reviewing this case, I would like to offer some suggestions to help optimise the results from ab74871.

1.) I agree with your customer that the band at 120kDa most probably is not in a complex since as your customer stated the samples are boiled and reduced. I can only speculate on the identity of this band, but would like your customer to do a no primary control to check if the secondary antibody is producing this band.

2.) I would like to suggest to decrease the boiling temperature to 65C and to prolong the heating time up to 10 min. Your target is a multi-pass membrane protein, and these proteins have the unfortunate tendency to aggregate during boiling periods with high temperatures and beta- mercaptoethanol due to their hydrophobic nature.

3.) Since the protein is expressed in the mitochondria, I can recommend to use a lysisbuffer that is a little harsher. When results are unpredictable, the detection is usually on its limits. Therefore it will work sometimes and sometimes not without apparent reasons. maybe a standard SDS lysisbuffer will help.

4.) Since rat is only a species that is predicted to react but not experimentally tested by us, I suggest to use a positive control that will deliver results like, for example, HeLa cell lysate.

We are happy to offer this technical support. In the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund.

I hope this information is helpful, thank you and your customer for your cooperation. Should the suggestions not improve the results, please do not hesitate to contact me again with the further requested details.

Answer

Thank you for your interest.

Mouse and rat are predicted species so it is likely that ab74871 will recognize them but w do not have any experimental data to support it.

I could certainly offer a testing discount for your customer. Would you be so kind to pass his/her contact details so that we can update our system.

I look forward to hearing from you soon.
