    • Function

      Calcium-binding protein. Has antimicrobial activity towards bacteria and fungi. Important for resistance to invasion by pathogenic bacteria. Up-regulates transcription of genes that are under the control of NF-kappa-B. Plays a role in the development of endotoxic shock in response to bacterial lipopolysaccharide (LPS) (By similarity). Promotes tubulin polymerization. Promotes phagocyte migration and infiltration of granulocytes at sites of wounding. Plays a role as pro-inflammatory mediator in acute and chronic inflammation and up-regulates the release of IL8 and cell-surface expression of ICAM1. Extracellular calprotectin binds to target cells and promotes apoptosis. Antimicrobial and proapoptotic activity is inhibited by zinc ions.

    • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human cervix carcinoma tissue labeling MRP8 with purified ab92331 at 1/500.

      Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9. ab97051, an HRP-conjugated goat anti-rabbit IgG (H+L) was used as the secondary antibody (1/500).

      Negative control using PBS instead of primary antibody. Counterstained with hematoxylin.

    • ab92331 (purified) at 1/30 immunoprecipitating MRP8 in HL-60 (Human promyelocytic leukemia cell line) cell lysate (Lane 1).

      Lane 2 - rabbit monoclonal IgG instead of ab92331 in HL-60 lysates. For western blotting, a peroxidase-conjugated goat anti-rabbit IgG (H+L) was used as the secondary antibody (1/1500).

    We have two lots in stock currently. I don't see a recentorder from Miami so unfortuantely I can't tell you exactly which lot you would get. But the current lots we have in stock are GR76267-2 at 0.2810 mg/ml and GR22309-5 at 0.2820 mg/ml.

    The reducing and denaturing conditions are most commonly used for antibodies, only handful of antibodies detects the native and non-reduced protein which is why this information is not necessary to be published on datasheet.

    As a rule of thumb if nothing is mentioned on the datasheet then it means the conditions are reduced and denaturing. In case of native form specificity, the information will be on the datasheet.

    ab92331 and ab92507 will be OK to use in reduced and denaturing conditions.

    As both antibodies contain Glycerol, they should be quite safe from Frost/Thaw damage (as they shouldn't freeze at all). In the case of these two antibodies the aliquots are rather a protection against contamination of the whole vial.

    I would like to reassure you that our antibodies have all been tested successfully and would be covered by our guarantee in the applications listed on the datasheets. Therefore, any MRP8 and MRP14 antibody tested in the species and application you are using should be suitable for your applications.

    Also, although we produce many in-house antibodies, we obtain other antibodies from a wide range of sources. Therefore, comparison experiments will not often have been done.

    If you wish to be more certain, I can suggest choosing the antibody that has been best characterized. By this I mean the antibody that has been tested in the most applications and has the most data available (on the datasheet).

    For your experiment, maybe an antibody that will bind to both proteins will be suitable? ab17050 recognises an epitope on the MRP8 + MRP14 complex which is not exposed on the individual subunits.
