  • Function

    Calcium-binding protein. Has antimicrobial activity towards bacteria and fungi. Important for resistance to invasion by pathogenic bacteria. Up-regulates transcription of genes that are under the control of NF-kappa-B. Plays a role in the development of endotoxic shock in response to bacterial lipopolysaccharide (LPS) (By similarity). Promotes tubulin polymerization. Promotes phagocyte migration and infiltration of granulocytes at sites of wounding. Plays a role as pro-inflammatory mediator in acute and chronic inflammation and up-regulates the release of IL8 and cell-surface expression of ICAM1. Extracellular calprotectin binds to target cells and promotes apoptosis. Antimicrobial and proapoptotic activity is inhibited by zinc ions.

  • IHC image of ab20220 staining in normal human tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab20220, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Thank you for your email. MRP-8 is Myeloid-related protein 8. MRP-8 and MRP-14 are cytosolic calcium-binding proteins of 8kDa and 14kDa that form a heterodimer. Thank you for your enquiry. IgM and IgG denotes the class of the antibody. Structurally antibodies are composed of one or more copies of a Y shaped unit. Each Y unit contains two identical heavy chains and two identical light chains. Antibodies are divided into 5 classes, IgG, IgM, IgA, IgE and IgD on the basis of the number of Y units and type of heavy chain polypeptide they contain. IgM is the first response antibody and IgG is the major Ig in serum. Ab20220 has been tested for application in Western blot, ELISA, Flow Cytometry/FACS and cross-reacts with human MRP8. I'm not aware of this antibody being used with PBMCs and would suggest using a positive control such as neutrophil lysate (particularly neutrophils from spleen). These two hyrbridomas were produced in the same fursion and so the immunogen was the same for both: Recombinant human MRP8 As to which may be better in rat, neither have been tested and so I don't know for sure if they would recognise rat MRP8. If it is a hightly conserved protein accress species it may be worth trying. As 7C12 was used in the original scientists publications, I would suggest that this one may be the stronger of the two. However we do not have any information about epitope so it may be the case that the other happens to recognise a more conserved region.
