MTR4, or superkiller viralicidic activity 2 like 2, may be involved in pre-mRNA splicing. MTR4 is an exosome-associated protein and may be involved in the recruitment of the exosome to pre-rRNA, to mediate the 3' end processing of 5.8S rRNA. -

Detection of human MTR4 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-MTR4 antibody ab70551 used for IP at 3 µg per reaction. MTR4 was also immunoprecipitated by rabbit anti-MTR4 antibody ab70552. For blotting immunoprecipitated MTR4, ab70551 was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

IHC image of ab70551 staining in human normal tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab70551, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
