Después de consultar este caso con mis compañeros del laboratorio, acerca de los siguientes anticuerpos contra Mu Opioid Receptor validados en tejidos de rata: ab10275, ab17934, ab64746, ab128013 y ab134054, me temo que ninguno de ellos ha demostrado reconocer el epítopo extracelular de la proteína.
Todos ellos se han generado a partir del extremo C terminal de la proteína, por lo que, a pesar de no haberse mapeado el epítopo que reconocen, probablemente detecten el dominio citoplásmico de la misma.
La solución que te propongo es testar uno de los anticuerpos que hay en el catálogo hecho a partir del extremo N terminal de la proteína, y testarlo en rata. Tenemos en abcam un sistema de evaluación de anticuerpos por el cual al testar uno de ellos en una aplicación o especie que no se ha testado, puedes beneficiarte de un descuento en tu próxima compra por valor del anticuerpo, si lo usas en esa especie o aplicación nueva y nos mandas los resultados a la web en forma de Abreview.

Thank you for taking the time to complete our questionnaire and contact us. I am sorry to hear the customer has repeatedly had difficulty obtaining satisfactory results from this antibody.

I appreciate the time spent in the laboratory and understand the concerns. It is regrettable the results have not been successful.

Having investigated and discussed this case with our immunohistochemistry experts, we figured out that the fixation of the fresh frozen mouse brain tissue sections might be the crucial step:

As indicated by the customer, a fixation with 4% PFA for 30 min at room temperature is performed. This step certainly results in the formation of methylene bridges, which cross-link proteins and therefore mask antigenic sites. PFA-fixed tissues thus require an antigen retrieval step before immunohistochemical staining can proceed. However, this procedure is rather unsuitable for delicate frozen brain sections, and we therefore recommend fixation with solvents like acetone, methanol or ethanol. The fixative should be pre-cooled and fixation carried out for 5-10 min at room temperature. We believe optimising this fixation step would improve the results significantly for both ab10275 and ab64746.

If this suggestion has not been tried already, we would appreciate if the customer can consider trying the optimisation. I hope you would agree that it would be preferable if we can get the experiment working and giving good results. Should the suggestion not improve the results, again we will be pleased to arrange an immediate credit note as requested.
