    • Function

      Coats the epithelia of the intestines, airways, and other mucus membrane-containing organs. Thought to provide a protective, lubricating barrier against particles and infectious agents at mucosal surfaces. Major constituent of both the inner and outer mucus layers of the colon and may play a role in excluding bacteria from the inner mucus layer.

      Colon, small intestine, colonic tumors, bronchus, cervix and gall bladder.

      Contains 1 CTCK (C-terminal cystine knot-like) domain.
      Contains 1 TIL (trypsin inhibitory-like) domain.

      modifications

      O-glycosylated.
      May undergo proteolytic cleavage in the outer mucus layer of the colon, contributing to the expanded volume and loose nature of this layer which allows for bacterial colonization in contrast to the inner mucus layer which is dense and devoid of bacteria.
      At low pH of 6 and under, undergoes autocatalytic cleavage in vitro in the N-terminal region of the fourth VWD domain. It is likely that this also occurs in vivo and is triggered by the low pH of the late secretory pathway.

        Ab7848 staining human normal colon. Staining is localised to the membrane.
        Left panel: with primary antibody diluted 1:4000. Right panel: isotype control.
        Sections were stained using an automated system DAKO Autostainer Plus , at room temperature. Sections were rehydrated and antigen retrieved with the Dako 3-in-1 AR buffer EDTA pH 9.0 in a DAKO PT Link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 minutes. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS), then incubated with primary antibody for 20 minutes, and detected with Dako Envision Flex amplification kit for 30 minutes. Colorimetric detection was completed with diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required.

      Question

      I have inquiries about your products, ab7848 and ab8605. A researcher is looking for MUC1 antibody and MUC2 antibody. Please let me know if the ab7848 should detect MUC2 in gastric mucosa and the ab8605 should detect MUC1 in intestinal mucosa, respectively. I can't find these descriptions in your datasheets.

      Answer

      Thank you for contacting us with your enquiry. The SwissProt database links on the datasheets for these two antibodies give some information on the tissue expression of Muc1 and muc2. Muc 2 (ab7848) is expressed in Colon, small intestine, colonic tumors, bronchus, cervix and gall bladder. Muc 1 (ab8605) is expressed on ductal epithelial cells and on activated T-cells. Aberrantly glycosylated forms expressed in human epithelial tumors, such as breast or ovarian cancer and also in non-epithelial tumor cells. I have also searched a reference which gives details of Muc 1 (and Muc 2)expression in normal and tumour gastric and intestinal tissue: J Clin Pathol. 1996 July; 49(7): 560–564. Significance of MUC1 and MUC2 mucin expression in colorectal cancer. Y Ajioka et al. I hope this information is helpful.
