Target

  • Relevance

    Mucins are high molecular weight glycoproteins with 80% carbohydrates and 20% core protein. Gastric Mucin 5AC antigen is found in columnar mucus cells of surface gastric epithelium and in goblet cells of the fetal and precancerous colon but not in normal colon. Resurgence of gastric mucin during colonic carcinogenesis is suggestive of either re-expression of the peptide core of gastric mucin in the adult colon or due to changes in the glycosylation pattern of mucin, which expose the hidden Mucin 5AC antigen.

  • Overlay histogram showing HeLa cells stained with ab218363 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Triton X-100 for 15 min. The cells were then incubated in 1x PBS / 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (ab218363, 1/50 dilution) for 30 min at 22°C.

    Isotype control antibody (black line) was Rabbit IgG (monoclonal) Alexa Fluor® 594 (ab208568) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.

    Acquisition of >5,000 events were collected using a 50 mW Yellow/Green laser (561nm) and 610/20 Bandpass filter.

    This antibody gave a positive signal in HeLa cells fixed with 4% formaldehyde (10 min)/permeabilized with 0.1% PBS-Triton X-100 for 15 min used under the same conditions.
