    E3 ubiquitin ligase. Regulates proteasomal degradation of cardiac troponin I/TNNI3 and probably of other sarcomeric-associated proteins. May play a role in striated muscle atrophy and hypertrophy by regulating an anti-hypertrophic PKC-mediated signaling pathway. May regulate the organization of myofibrils through TTN in muscle cells.

  • ab4125 at 1.25 µg/ml staining human skeletal muscle by IHC-P.  Tissue sections were subjected to steamed antigen retrieval with citrate buffer (pH6), AP-staining.  Shows strong staining in rare nuclei, and weak staining of muscle fibres in longitudinal section.

  • ab4125 staining MURF1 in mouse lung tissue section by Immunohistochemistry (Frozen sections). Tissue underwent fixation in paraformaldehyde and blocked in 1%BSA for 10 minutes at 20°C. The primary antibody was diluted 1/200 and incubated with sample for 20 hours at 20°C. A Biotin conjugated rabbit polyclonal to goat IgG, diluted 1/500 was used as secondary.

As promised, I have contacted previous academic customers that have purchased this product in order to ascertain whether they have been using this product in Western blotting or not. It is now a case of waiting to see whether I receive any responses. I have not forwarded your contact details to our customers as I do not wish customers to feel pressured in any way. Thank you for your message below. I checked the sequence of rat MURF1 and it isn't totally identical to the human immunogen sequence we used. Therefore the antibody may yet work in human samples (although we have had not positive feedback on this yet) but not in rat. I am afraid I don't have any further feedback to give.

Ab4125 is expected to cross-react with Human due to sequence homology but has not yet been tested in other species. Ab4125 is a fast track antibody and is not yet fully characterised; it is shown by ELISA to have high titre values against the immunogen and is currently being tested for use in Western blotting.
