The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
This product gave a positive signal in Jurkat cells fixed with 80% methanol (5 min).
FunctionRegulates actin polymerization by stimulating the actin-nucleating activity of the Arp2/3 complex. Binds to HSF1/HSTF1 and forms a complex on heat shock promoter elements (HSE) that negatively regulates HSP90 expression.
Sequence similaritiesContains 1 CRIB domain.
Contains 1 WH1 domain.
Contains 2 WH2 domains.
Cellular localizationCytoplasm > cytoskeleton. Nucleus. Preferentially localized in the cytoplasm when phosphorylated and in the nucleus when unphosphorylated.

ab208524 staining N WASP in Jurkat cells. The cells were fixed with 80% methanol (5 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab208524 at 1/100 dilution (pseudocolored in green) and
ab195884, Rat monoclonal to Tubulin (Alexa Fluor® 647), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
