Number of tests vary depending on platform used in the assay. The probe contains sufficient material for either 100 assays in a standard 96 well plate or 10 assays in an imaging mini dish 35mm.
Probe Preparation and Cell Loading
Probe is supplied as 100 µg aliquots in plastic vials.
1. Add 100 µL of water or medium to the vial and vortex for 30 seconds to produce 1 mg/mL stock solution of the probe.
2. For use with cultured cells, spheroids or tissue slices dilute probe stock 1:100 with medium (10 µg/mL working concentration) and add this solution to the cells.
3. Incubate at 37°C in a CO2 incubator for 3-24h, then wash the cells 1-2 times with fresh medium and conduct optical measurements.
Depending on the cell type and application, optimal probe concentration (typical range 2- 20 µg/mL) and loading time may vary.
Temperature and gas equilibration of samples during the measurement is important since the probe is sensitive to both O2 and T.
