Specifications

Our Abpromise guarantee covers the use of ab77930 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

  • Applications

    SDS-PAGE

    Western blot

  • Form

    Lyophilised
  • Additional notes

All human source materials have tested negative for HIV-1 and HIV-2 and non-reactive with HCV and HBc antibodies and HBsAg. No test guarantees a product to be non-infectious. Therefore, all material derived from human fluids or tissues should be considered as potentially infectious.

Storage Notes: Prolonged exposure to light will degrade the ceruloplasmin. This will lead to a decrease in the blue color of the product.

  • Function

Ceruloplasmin is a blue, copper-binding (6-7 atoms per molecule) glycoprotein. It has ferroxidase activity oxidizing Fe(2+) to Fe(3+) without releasing radical oxygen species. It is involved in iron transport across the cell membrane.
  • Tissue specificity

Expressed by the liver and secreted in plasma.
  • Involvement in disease

Defects in CP are the cause of aceruloplasminemia (ACERULOP) [MIM:604290]. It is an autosomal recessive disorder of iron metabolism characterized by iron accumulation in the brain as well as visceral organs. Clinical features consist of the triad of retinal degeneration, diabetes mellitus and neurological disturbances.
Note=Ceruloplasmin levels are decreased in Wilson disease, in which copper cannot be incorporated into ceruloplasmin in liver because of defects in the copper-transporting ATPase 2.
  • Sequence similarities

Belongs to the multicopper oxidase family.
    Contains 3 F5/8 type A domains.
    Contains 6 plastocyanin-like domains.
  • Cellular localization

    Secreted.
  • Information by UniProt

Answer

For ab77930 Human Ceruloplasmin protein, the copper ions are stably bound as long as you do not change the buffer. The buffer this was lyophilized from is as follows:

Buffer: Lyophilized from 50 mM Potassium Phosphate, pH 6.8, 100 mM Potassium Chloride, 20 mM E-amino-n-caproic acid and 5 mM EDTA.
NOTE: Exposure to sodium (in the form of sodium chloride, sodium phosphate, sodium azide as well as other sodium containing reagents) should be avoided, as ceruloplasmin may precipitate under these conditions. Buffers that ceruloplasmin is exposed to should be pH adjusted with potassium hydroxide.

Thank you for contacting us.
We unfortunately haven't tested this product for its enzymatic activity so I am sorry we are unable to confirm that the product can act as enzyme. The activity might have to be determined by end user.
This product is recommended to be used in SDS-PAGE and WB and these applications does not involve enzymatic activity.
I hope this information is nevertheless helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Answer

Thank you for your new enquiry regarding ab77930. GR44819 is the same as batch: 1104116: Please reconstitute a 1mg vial with 310uL of water.  This will return the product to the original protein concentration of 3.23mg/mL and to the original buffer formulation. Hope this will be useful for you and your customer.

Answer

Thank you for your enquiry regarding ab77930. The concentration before lyophilization is lot-specific (each lot can have a different concentration). The label of the vial should indicate the actual concentration. Batch 699123: Reconstitute a 1mg vial with 167.1uL of water. This will return the product to the original protein concentration of 5.98mg/mL and to the original buffer formulation. Batch 887371 and 855379: Reconstitute a 1mg vial with 436.7uL of water. This will return the product to the original protein concentration of 2.29mg/mL and to the original buffer formulation. Batch 1104116: Reconstitute a 1mg vial with 310uL of water. This will return the product to the original protein concentration of 3.23mg/mL and to the original buffer formulation. If your customer chooses to reconstitute the vial using 1mL of water, this will result in a protein concentration of 1mg/mL, but the buffer formulation in the final reconstituted solution will be diluted.   I hope this helps and if I can assist further, please do not hesitate to contact me.
