    Nuclear hormone receptor. The steroid hormones and their receptors are involved in the regulation of eukaryotic gene expression and affect cellular proliferation and differentiation in target tissues. Can activate the transcriptional activity of TFF1.

      ab79413 staining Estrogen Receptor alpha in Baboon placenta and fetal ovary tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 5% serum for 30 minutes at 22°C; antigen retrieval was by heat mediation in 10mM sodium citrate, pH 6.0. Samples were incubated with primary antibody (1/500 in PBS + 5% normal goat serum) for 16 hours at 4°C. A Biotin-conjugated goat anti-rabbit IgG polyclonal (7µg/ml) was used as the secondary antibody.

    thank you for contacting us. I am currently looking into wether we have any suitable antibodies for the specifications you have provided. We have a number of antibodies for which the immunogen used is within the C-terminal region of human Estrogen Receptor alpha (ab21232, ab15284. ab56177, ab1001499, ab79413 and ab45005 as you already have identified). However, in order to make sure that the immunogen used for each of these antibodies lies within your region of interest (residues 302-522) I am currently checking with the sources of these antibodies. I will also look into wether the two phospho specific antibodies we have (ab39222 and ab61049) may be suitable for your application. As soon as I have further information I will let you know.

    Dear Sir/Madam, I would like to introduce a human mutant form of ERα (S554fs, which carries a frameshift mutation in aa 554) in an ERa-positive mouse cell line. It is very important for my experimental design to have a human-specific antibody, that would not cross-react with the endogenous, mouse wild type ERα in Western blots. Failure to do so, would result in false positive results. Are the epitopes of your human-specific ERa antibody, catalogue number ab79413, upstream aa 554? I would greatly appreciate your response to my query at your earliest convenience, so that I can proceed to the purchase of the most suitable ERα antibody. Yours faithfully

    Answer

    Thank you for your enquiry. I can confirm that the immunogen for ab79413 Estrogen Receptor alpha antibody [EPR703(2)] is not upstream of amino acid residue 554. It is specific to the C-terminus, ending at amino acid 594. Regrettably, I would suggest this antibody would not be suitable for your experiments, since it would not recognize the mutant ERα.
