    Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human kidney tissue labelling Urokinase with purified ab133563 at 1/150. Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9. A prediluted HRP-polymer conjugated anti-rabbit IgG was used as the secondary antibody. Negative control using PBS instead of primary antibody. Counterstained with Hematoxylin.

Question

Urokinase is synthesized as a zymogen form (app. 54 kDa), and is activated by proteolytic cleavage. The two resulting chains are kepttogether by a disulfide bond. So when i reduce my samples for western blotting I cleave the disulfide band and get two bands of app 18 and 33 kDa. So why do you only get one band of app. 49 kDa with this antibody. Shouldn´t I get two bands; the zymogen form and one of the smaller bands (18 or 33 kDa, depending on where the epitope is)?

Answer

According to the literature, the ˜54/49kD can be detected under reducing conditions and it could represent the single chain pro-uPA. Also, looking at some of the literatures’ data, the detection of the pro-uPA, 18 and 33kD bands will vary from one sample to another.

Furthermore, it’s possible that the reducing agent could make a difference too. For example, to test this antibody, we used BME, which is a weaker reducing agent than DTT and so this may account for the single band at 49 kDa being observed.
