      CNAP1/hCAP-D2 is a regulatory subunit of the condensin complex, a complex required for conversion of interphase chromatin into mitotic-like condensed chromosomes. The condensin complex probably introduces positive supercoils into relaxed DNA in the presence of type I topoisomerases and converts nicked DNA into positive knotted forms in the presence of type II topoisomerases. It may target the condensin complex to DNA via its C-terminal domain (referenced from Swissprot).

Paraffin embedded human testis tissue was incubated with ab34338 (1/100 dilution) for 30 mins at room temperature. Antigen retrieval was performed by heat induction in citrate buffer pH 6.

    Thank you for confirming the details. The antibodies should be working fine with the protocol details you have provided. I am not sure why these antibodies fail. I am happy in sending replacement vials of both of these antibodies. Could you let me know how you would like to proceed? I would also like to draw your attention for storing these antibodies please store at -20C after liquating these and in case you need to keep them 4C please try to use aliquots within 1-2 weeks.     Thank you for your enquiry regarding ab4448 and for taking the time to provide some useful details of the experiments. I am very sorry to hear that you are having problems with this antibody. Ab4448 is a very high characterized antibody which has been used in tens of publications and Abreview; failing of this antibody actually surprises us. Even ab34338 was tested successfully with human lysates so this antibody should also be fine with human cells.   We unfortunately can’t guarantee the use of these antibodies for Dog species. I have read the protocol which looks fine to me however I would like to ask few questions which would help me identify the source of the problem; - Could you provide details e.g. at what temperature the antibodies were stored? - We have used Hela cells to test these antibodies; this is a easily available cell line which I would recommend using as a positive control. - You have mentioned using Tubulin as positive control. How the tubulin staining was? - Does these antibodies fail to give staining at all or giving high background? - For how long the cells were permeabilized. - Have you tried same procedure successfully before? - Why the reporting of problem was delayed for 8 months.
