  • Species reactivity

    Reacts with: Human
  • Immunogen

Synthetic peptide corresponding to Human NCOR2/SMRT aa 994-1005.

    (Peptide available as ab4910)

  • Positive control

    • HeLa cell extract
  • General notes

In ChIP ab2780 gives a maximum at 60 min (more than 60%) whereas ab2781 gives a minimum (around 5%) at this time and ab 2780 gives a minimum (less than 5%) at 30 min whereas ab2781 gives a maximum (50%). This protein is corepressor of the nuclear hormone receptors and is not supposed to be recruited in presence of the hormone (here oestrogen), and should be maximal before induction, which is not the case here for both antibodies!

    Previously labelled as NCOR2.

Our Abpromise guarantee covers the use of ab2781 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ChIP Use at an assay dependent concentration.
IHC-Fr Use at an assay dependent concentration.
WB Use a concentration of 4 µg/ml. Detects a band of approximately 250, 220 kDa (predicted molecular weight: 274 kDa).Can be blocked with Retinoic Acid Receptor alpha peptide (ab4910).
Flow Cyt Use 2µg for 106 cells.

ab170190 - Mouse monoclonal IgG1, is suitable for use as an isotype control with this antibody.

IP Use at an assay dependent concentration.
IHC-P Use at an assay dependent concentration.

  • Function

Transcriptional corepressor of NR4A2/NURR1 and acts through histone deacetylases (HDACs) to keep promoters of NR4A2/NURR1 target genes in a repressed deacetylated state (By similarity). Mediates the transcriptional repression activity of some nuclear receptors by promoting chromatin condensation, thus preventing access of the basal transcription. Isoform 1 and isoform 5 have different affinities for different nuclear receptors.
  • Tissue specificity

Ubiquitous. High levels of expression are detected in lung, spleen and brain.
  • Sequence similarities

Belongs to the N-CoR nuclear receptor corepressors family.
    Contains 2 SANT domains.
  • Domain

The N-terminal region contains repression functions that are divided into three independent repression domains (RD1, RD2 and RD3). The C-terminal region contains the nuclear receptor-interacting domains that are divided in two separate interaction domains (ID1 and ID2).
The two interaction domains (ID) contain a conserved sequence referred to as the CORNR box. This motif is required and sufficient to permit binding to unligated TR and RARS. Sequences flanking the CORNR box determine nuclear hormone receptor specificity.

  • Overlay histogram showing HeLa cells stained with ab2781 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab2781, 2µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

  • All lanes : Anti-NCOR2/SMRT antibody [1542] (ab2781) at 1/100 dilution

Lane 1 : HeLa cell lysate
    Lane 2 : Neuro-2a cell lysate

Lysates/proteins at 25 µg per lane.

    Predicted band size: 274 kDa
    Observed band size: 220,250 kDa
why is the actual band size different from the predicted?

Customer reviews and Q&As

Thank you for your enquiry. From my understanding, NCoR2 and SMRT are the same thing. When you look at the SwissProt database entry for NCoR2 (primary accession number Q9Y618), a synonym is SMRT. We do have antibodies to NCoR (thyroid hormone- and retinoic acid receptor-associated corepressor 1) - ab3482 and ab14591. Please let me know if you need additional assistance.

Both monoclonal products (ab2781 and ab2780) to NCoR2 have the same immunogen: Synthetic Peptide: C G G A(994) S A S D P H R E K T Q(1005). There really is no difference except for the clone. Ab2780 = Cone 1212; Ab2781 = Clone 1542. Unfortunately, we don't have any information regarding the cross-reactivity of these antibodies with I do not have any information as to their cross-reactivity with NcoR1. If you have any additional questions, please contact us again.

Thank you for your enquiry. Ab2781 has been tested for application in ChIP. However, the antibody has only so far been tested for cross-reactivity with human and has not yet been tested for cross-reactivity with mouse.
