Thank you very much for your email.
The recent lots developed for this ab are stable at even -20C so we changed the recommended conditions on datasheet last year in March 2012. This change however does not affect the stability of the previous batches including 928905 which was recomended to be stored at 4C. The antibody customer received was absolutely fine.
We obviously cannot refund the vial bought in 2010 because it is out of Abpromise guarantee we however have decide to give this customer a 50% credit note. Please tell the customer to buy ab16912 and once they test it and are satisfied I will refund the price they paid for original invoice dated Oct 2010. In case the recent batch fails we will refund the current vial, as well as 50% amount of original vial. Please ask customer to submit an Abreview for extra perks.

General Information
Antibody storage conditions (temperature/reconstitution etc): 4 degrees
Description of the problem (high background, low signal, no signal etc.):
Doesn’t recognize the human Nectin2, although does recognize the mouse orthologue.
Sample (Species/Cell type/Cell line etc.): 721.221 parental cells, 721.221 transfected to express mouse Nectin2 fused to HA tag or mouse Nectin2 without a tag, 721.221 transfected to express human Nectin2, 293T and A549 (previously stained with anti-human Nectin2 clone TX31 BioLegend cat# 337401).
Sample preparation for single cell preparation (Buffer etc.):
FACS buffer (0.5% BSA, 0.025% NaN3 in PBSx1).
Number of cells used: 100,000 per stain.
Permeabilization, fixation step: no permeabilization or fixation.
Blocking conditions (Buffer/time period, Blocking agent etc.): no blocking.
Primary Antibody (Manufacturer/Species/Diluent/Dilution/Incubation time, Wash step): 2ul (=0.2ug) antibody in 100ul FACS buffer for 1h on ice. Washed and incubated with secondary.
Secondary Antibody (Manufacturer/Species/Diluent/Dilution/Incubation time, Wash step):
APC-conjugated F(ab’)2 anti-Rat IgG Jackson cat# 712-136-153 diluted 1:200 in 100ul FACS buffer for 30 min on ice. Washed twice with FACS buffer.
Positive and negative controls used (please specify): staining of 721.221 parental cells is the negative control, while 721.221/mouse Nectin2 transfectants, 293T and A549 are the positive controls.
Detection method: FACS analysis
Optimization attempts (problem solving):
How many times have you tried the FACS? Twice.
Have you run a "No Primary" control? The background is always the staining with secondary only.
Do you obtain the same results every time? yes
What steps have you altered? Since mouse nectin2 was detected by this Ab, I assume the staining procedure and reagents are good and don’t need to be altered.
Additional Notes:
Attached is previous staining of 293T, A549 and 721.221/human Nectin2 transfectants with anti-human Nectin2 clone TX31 BioLegend cat# 337401 Ab and the recent staining with the antibody discussed here. It should be emphasized that this particular antibody was purchased for its ability to detect both human and mouse Nectin2 proteins.
