    ELISA: 1/2000 - 1/10000.
    WB: 1/1000. Detects a band of approximately 9.7 kDa.

    Not tested in other applications.
    Optimal dilutions/concentrations should be determined by the end user.

    This antibody using the specified conditions may recognize other prominent intrinsic bands (UBLs or conjugates). Other intrinsic bands are readily detectable at lower dilutions.

    • Function

      Ubiquitin-like protein which plays an important role in cell cycle control and embryogenesis. Covalent attachment to its substrates requires prior activation by the E1 complex UBE1C-APPBP1 and linkage to the E2 enzyme UBE2M. Attachment of NEDD8 to cullins activates their associated E3 ubiquitin ligase activity, and thus promotes polyubiquitination and proteasomal degradation of cyclins and other regulatory proteins.

      ab4751 tested by immunoblot against a yeast cell lysate.  A dilution of the antibody between 1:200 and 1:1,000 will show strong reactivity specifically with free Rub1 protein (indicated by arrow) and Rub1 conjugates.  In this blot the antibody was used at a 1:500 dilution incubated overnight at 4° C in 5% non-fat dry milk in TTBS.   Detection occurred using a 1:2000 dilution of Donkey polyclonal to Rabbit IgG (HRP) (ab7083) for 1 hour at room temperature.  A chemiluminescence system was used for signal detection (Roche). ab4751 tested by immunoblot against a yeast cell lysate.       Immunoblot of Rub1 fusion protein.  Anti-Rub1 antibody generated by immunization with recombinant yeast Rub1 was tested by immunoblot against yeast lysates expressing the Rub1-GFP fusion protein and other UBL fusion proteins.
