      Question

      Thank you for your response. I apologize for the delay in getting back to you, but I wanted my lab to double check that the antibody staining we had was correct - and unfortunately it appears that the Negr1 antibody (ab133676, lot number GR96807-2) is not working for us in western blots. I have attached a PDF showing a western blot using the Abcam antibody side by side with a Sigma antibody. It appears that our knockdown of Negr1 reduces the levels of the protein when probed with a Sigma antibody, but when we use the Abcam antibody. These westerns were performed with the same lysates. We have done this a few times with similar results. We also in parallel conducted RT-qPCR to ensure our knockdown, and as you can see our knockdown is very efficient.

      If possible, I would appreciate as a replacement to get the Anti-MARK2 antibody [EPR8553] (ab133724) from Abcam.

      Please let me know if this is possible. Thank you very much for your help.

      Question

      We recently bought the abcam NEGR1 antibody (ab133676, lot number GR96807-2) to use for some of our experiments and have been testing it by western blot. Unfortunately the antibody does not work as stated. Using the recommended protocols on the data sheet the antibody recognizes a band that runs at the appropriate size on a SDS-PAGE gel, but the band is not NEGR1. We have confirmed this by knocking down NEGR1, as we see no change in the band by western blot using the abcam antibody. We have confirmed knockdown using two methods: qPCR analysis which shows approximately 90% knockdown of mRNA, and western blot using antibody purchased from Sigma which shows a strong reduction in the NEGR1 protein. We would be happy to share the data with you. We were hoping to receive a refund or exchange for a different antibody as stated on the Abpromise guarantee. Please let me know what kind of information you might need.

      Would you be able to share some of your data with us (i.e. images of the Abcam antibody alongside the Sigma antibody)? This is one of our Epitomics products and our RabMAbs are intended to be highly specific, so I'll have to forward the data to our lab to have them take a look and reconsider some of the QA or whether or not to discontinue the ab.
