IHC-P Use a concentration of 1 - 5 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

    Seems to play a role in the development or function of the kidney glomerular filtration barrier. Regulates glomerular vascular permeability. May anchor the podocyte slit diaphragm to the actin cytoskeleton. Plays a role in skeletal muscle formation through regulation of myoblast fusion.

    Defects in NPHS1 are the cause of nephrotic syndrome type 1 (NPHS1) [MIM:256300]; also known as Finnish congenital nephrosis (CNF). A renal disease characterized clinically by proteinuria, hypoalbuminemia, hyperlipidemia, and edema. Kidney biopsies show non-specific histologic changes such as focal segmental glomerulosclerosis and diffuse mesangial proliferation. Some affected individuals have an inherited steroid-resistant form and progress to end-stage renal failure.

  • IHC image of Nephrin staining in a formalin fixed, paraffin embedded human normal kidney tissue section*, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with 85379 at 1 µg/ml for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

I am sorry to hear that these antibodies are not working as expected.
As for ab58968, we had replaced this antibody free of charge with ab85379. Since ab85379 was not tested in rat, sheep, and pig tissue, I had sent you a testing discount code. The code (xxx) is still valid until its expiration date of xxx.
Which species did you try ab85379 on?
As for ab50339, it has only been tested in WB, IHC-Fr, ICC/IF, but not paraffin embedded sections. It might work on paraffin sections, but we have no data to confirm this as it was not yet tested.
I'd be happy, though, to give you a testing discount code for trying it in IHC-P. Would you use it on the same species as ab85379?
