We have the Anti-Nestin antibody (10C2) of Abcam.Its catalog No# is ab22035.In its datasheet it is given that it can be used for IHC of paraffin embedded tissue sections. But it is given in datasheet to use either rhodamine or fluorescein conjugated anti-mouse secondary antibody.So can I use HRP conjugated secondary antibody provided with the EXPOSE kit of Abcam??? Is it necessary that I have to specifically use Flourescein/Rhodamine conjugated secondary antibody for its detection?? If so, can you please provide the protocol for Immunofluoresecnce in formalin fixed paraffin embedded tissue???

I can confirm thatthis antibody can be used with HRP conjugated secondary antibody without any problems.
The note on the datasheet is in reference on how it has been tested: the ab22035 has been tested on paraffin embedded tissue slides witha secondary antibody conjugated to FITC or Rhodamine. Inthat case, adilution of the ab22035 at1/100 to 1/200 gave good signal.
I would expect therefore, that using this antibody at 1/100 to 1/200 will also give good results using a secondary antibody coupled to HRP or the EXPOSE kit.
