    Netrins control guidance of CNS commissural axons and peripheral motor axons. Its association with either DCC or some UNC5 receptors will lead to axon attraction or repulsion, respectively. It also serve as a survival factor via its association with its receptors which prevent the initiation of apoptosis. Involved in tumorigenesis by regulating apoptosis.

I have attached the Abcam protocols books that contains some good IHC-P protocols, which contains some good general starting protocols. Specifically for ab39370, we recommend a starting primary antibody concentration of 1/100 - 1/200 and for ab77949, using a primary antibody concentration of 2.5 µg/ml. Although you may need to optimize these for your own conditions.

I am sorry to hear that you have been experiencing difficulties using this ab39370 anti-netrin1 antibody. Please find the protocols used for the ICC/IF results below.



1. Draw of culture medium with aspirator and add 1 ml of 3.7 % formalin in PBS solution to the dish. Let sit at room temp for 1 minute.



2. Aspirate formalin/PBS and add 1ml of cold methanol (-20°C, kept in well sealed bottle in fridge). Let sit for no more than 1 minute.



3. Aspirate methanol and add 1ml of PBS, making sure the specimen does not dry out. To block nonspecific antibody binding add ˜10ml of 1% goat serum, and incubate for 30 minutes. Then, add antibody reagents. Incubate for 1 hour at room temperature (37°C for 30 minutes to 1 hour, or 4°C overnight).



4. Remove primary antibody and replace with 1 ml of PBS. Let sit for 5-10 minutes, replace PBS and repeat twice, to give three washes in PBS.



5. Add 0.5 mls of secondary antibody. These are fluorescently labeled Goat anti mouse or rabbit antibodies and are conjugated to ALEXA dyes and are from Molecular probes (Eugene Oregon, the ALEXA dyes are sulphonated rhodamine compounds and are much more stable to UV than FITC, TRITC, Texas red etc.). Typically make 1:2,000 dilutions of these secondaries in PBS plus 1% goat serum, BSA or non fat milk carrier. Incubate for 1 hour at room temperature ( 37°C for 30 minutes to 1 hour, or 4°C overnight).



6. Remove secondary antibody and replace with 1 ml of PBS. Let sit for 5-10 minutes, replace PBS and repeat twice, to give three washes in PBS.



7. Drop on one drop of Fisher mounting medium onto dish and apply 22mm square coverslip. View under the microscope.

Thank you for your enquiry. I have contacted the originator of the product but unfortunately, the exact peptide sequence information is proprietary and cannot be released. However, I did confirm that it is a synthetic peptide corresponding to a region near the center of the protein [between residues 300 and 400 a.a. of Human sequence (O95631)]. This region contains a "laminin EGF-like 1" domain and is VERY highly conserved among vertebrates, being 100% conserved in the human, mouse, rat, xenopus laevis, and danio rerio gene products (although it isn't conserved with the Drosophila homolog). Also, the peptide length is around 12-20 amino acids.
