RelevanceNeurexin 2 alpha belongs to the Neurexin family, a group of neuronal proteins that function as cell adhesion molecules during synaptogenesis and intracellular signaling. Mammalian genomes contain just three neurexin genes. However, many (>200) neurexin isoforms may be expressed through the use of two alternative promoters and alternative splicing at up to five different positions.

ab34245 staining Neurexin II alpha protein in normal adult human cerebral cortex. Brown color indicates presence of protein, blue color shows cell nuclei. Paraffin embedded human cerebral cortex tissue was incubated with ab34245 (1/700 dilution) for 30 mins at room temperature. Antigen retrieval was performed by heat induction in citrate buffer pH 6.

Samples were blocked using 0.2% casein for 1 hour at 37°C.
ICC/IF image of ab34245 stained SKNSH cells. The cells were 4% formaldehdye fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab34245, 1µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
