      Thank you for contacting Abcam.

      I am sorry that ab109236 is not producing a strong 25 kDa band in Western blot for your mouse brain homogenate. As we discussed on the phone, I am sending you a positive control free of charge. I also recommend attempting a primary dilution of 1/500 in case your samples simply contain a low amount of Neurogenin 2. Protease and phosphatase inhibitors should also be added if you have not already done so. Additionally, since the Neurogenin 2 protein is localized to the nucleus we recommend using RIPA buffer.

      We initially discussed sending a HepG2 (liver carcinoma) cell lysate as the positive control, but after doing further searching I found a U-87 MG Nuclear Lysate (ab14903: human glioblastoma-astrocytoma) that has been used as a positive control for Neurogenin 2 and is more relavant to your research being a brain cell line as you desired.
