Application Abreviews Notes
IHC-P 1/1000. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
WB 1/500 - 1/1000. Detects a band of approximately 47 kDa (predicted molecular weight: 47 kDa). A doublet may be seen in some cell lines. For optimal results, primary antibody incubations should be performed at room temperature (see recommended protocol). The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background.

Thank you for your email. I am sorry to hear that you have been experiencing problems with this antibody.

I have read the details you have kindly provided and surprised to hear that 2 vials of this antibody have been failed. We have sold many vials of this batch without receiving a single complaint so protocol troubleshooting might help in this case. This is a rabbit polyclonal antibody where batch to batch variations are quite obvious; this is due to the way, rabbit's immune system responds to antigen so we always recommend our customers trying troubleshooting tips e.g. increasing or decreasing dilution of primary as well as secondary ab, increasing or decreasing the amount of loaded lysates etc

The following troubleshooting tips can be tried:

- Try primary antibody at 1/500 dilution; overnight incubation
- Try overnight incubation
- . Though previous batches gave no background staining with 5% milk however as observed form the image 5% BSA might improve blocking with this lot. Please use 5% BSA for blocking
