Samples were incubated with primary antibody (1/2500 in TBS/BSA/azide) for 16 hours at 21°C. A Biotin-conjugated Goat anti-rabbit IgG polyclonal (1/250) was used as the secondary antibody.

Thank you for contacting us yesterday in regards to the Anti-Nicastrin antibody (ab45425) and Anti-PEN2 antibody (ab109619).

As discussed over the phone, I have requested further information regarding the immunogen used to raise the two antibodies. Unfortunately the exact immunogen sequence of these antibodies is proprietary, however I can say that theanti-Nicastrin antibody (ab45425) has been raised against an 18 amino acidsynthetic peptide taken from within the region 351-400 ofhumanNicastrin (NP_056146).

As stated on the datasheet of the anti-PEN2 antibody (ab109619), this antibody has been raised against a 13 amino acid peptide taken from the N-terminus of the protein. Unfortunately I cannot provide any further information at this time as it is proprietary information.

However, in each case, if you could let me know the regions of your mutations and what you are trying to understand (the likelihood of cross reactivity?) I can pass this on and provide you with some further answers.

Thank you for your patience. I have received a reply from the originator of ab45425 (Nicastrin antibody). The immunogenic peptide sequence is proprietary information but they have performed a sequence alignment for you. Based on the alignment results, we know it is unlikely that this product will react with Dictyostelium discoideum and we do not recommend using ab45425 with this species.
