Target
-
Function
After binding acetylcholine, the AChR responds by an extensive change in conformation that affects all subunits and leads to opening of an ion-conducting channel across the plasma membrane. -
Sequence similarities
Belongs to the ligand-gated ion channel (TC 1.A.9) family. Acetylcholine receptor (TC 1.A.9.1) subfamily. Alpha-3/CHRNA3 sub-subfamily. -
Cellular localization
Cell junction > synapse > postsynaptic cell membrane. Cell membrane. - Information by UniProt

Anti-Nicotinic Acetylcholine Receptor alpha 3/CHRNA3 antibody (ab110801) at 1 µg/ml + Human spinal cord tissue lysate - total protein (ab29188) at 10 µg
Secondary
Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/10000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 57 kDa
Observed band size: 57 kDa
Additional bands at: 51 kDa (possible cleavage fragment)
Exposure time: 8 minutesThe band observed at 51 kDa could potentially be a cleaved form of v due to the presence of a 31 amino acid signal peptide. This blot was produced using a 10% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab110801 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
