Target

  • Function

After binding acetylcholine, the AChR responds by an extensive change in conformation that affects all subunits and leads to opening of an ion-conducting channel across the plasma membrane.
  • Involvement in disease

Defects in CHRNA4 are the cause of nocturnal frontal lobe epilepsy type 1 (ENFL1) [MIM:600513]; also symbolized ADNFLE. ENFL1 is an autosomal dominant epilepsy characterized by nocturnal seizures with hyperkinetic automatisms and poorly organized stereotyped movements.
  • Sequence similarities

Belongs to the ligand-gated ion channel (TC 1.A.9) family. Acetylcholine receptor (TC 1.A.9.1) subfamily. Alpha-4/CHRNA4 sub-subfamily.
  • Cellular localization

Cell junction > synapse > postsynaptic cell membrane. Cell membrane.

ab41172 (1/250 dilution) staining Nicotinic Acetylcholine Receptor alpha 4/CHRNA4 in human cerebellum using an automated system (DAKO Autostainer Plus). Using this protocol there is strong staining of nuclear and membrane compartment within the glia region of cells
Sections were rehydrated and antigen retrieved with the Dako 3 in 1 AR buffer EDTA pH 9.0 in a DAKO PT link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 mins. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 min and detected with Dako envision flex amplification kit for 30 minutes. Colorimetric detection was completed with Diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that, for manual staining, optimization of primary antibody concentration and incubation time is recommended. Signal amplification may be required.

Question

Dear xxxx,

As far as I know, unfortunately there is no data about the protein sequences of the Helix pomatia acetylcholine receptor alpha 4 and 7. Even so I hope I can test one of the antibodies.

Best regards,

Answer

Thank you for your reply.

As we have no sequence for the proteins from Helix pomatia I cannot predict how likely the antibodies you would like to try will cross react. We would therefore not usually offer the testing discount scheme for this. However I am willing to make an exception in this case if you would still like to try it. The testing discount codes for the two antibodies as follows:

ab10096, discount code: xxxxxx, expiration date: 13th March 2013, value: €380.00

ab41172, discount code: xxxxxx, expiration date: 13th March 2013, value: €370.00

The codes above will give you €380.00 and €370.00 respectively off your next order before the expiration date. To redeem this offer, please submit an Abreview for Helix pomatia in Western blotting and include this code in the “Additional Comments” section so we know the Abreview is for this promotion. Please remember that submission of the Abreview is sufficient for the discount code to become active.
For more information on how to submit an Abreview, please visit the site:

www.abcam.com/Abreviews

Remember, we publish both positive and negative Abreviews on our datasheets so please submit the results of your tests. The code will be active once the Abreview has been submitted and can be redeemed in one of the following ways:

1) Call to place your order and mention the code to our customer service department;
2) Include the code in your fax order;
3) Place your order on the web and enter the promotional code.

Any feedback that you can provide will be greatly appreciated, whether positive or negative. If you have any further questions, please do not hesitate to contact us. We look forward to receiving your Abreview and wish you luck with your research.

Question

Thanks a lot for your answer it was really fast!
We do agree to order and get discount for the antibodies you have suggested:

CDC42 Binding Protein Kinase Beta antibody ab61328

CNKR2 antibody ab64844

NEK7 antibody ab80948

MEK3 antibody ab5428

BMX antibody ab73887

Muscarinic Acetylcholine Receptor 1 antibody ab75178

PPP4C antibody sb70888
But I have one more question.
On the https://www.abcam.com/index.html?pageconfig=resource&rid=11998 of your site in FAQ part, the last question isreferred to the antibodies that have already been tested in the applications for some species, but have no image. And the answer says that there is a possibility to get discount also for those antibodies.
There are still 11 antibodies that have no images for the proposed species. if it is possible to give us some more discounts, we will submit the reviews including images for the proposed antibodies.
Can you please have a look one more time?

Thank you!
Best regards

For the antibodies we can provide a discount for, I can offer a discount off a future purchase if you buy the antibody(ies) now, test them in mouse, rat or both as indicated above, and submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount provided for each individual antibody would be to the value of 1 free primary antibody.

If you are interested in this offer, please follow these steps:

1. Reply to this e-mail to let me know that you would like to proceed and test these antibodies in mouse, rat or both. I will then send a discount code. This code must be issued before purchasing the antibodies so please wait for my reply before ordering.

2. Purchase the antibodies either by phone, fax, or online (www.abcam.com).

3. Test it in mouse, rat or both.

4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews.

5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any [PRIMARY ANTIBODY /PROTEIN] ordered and the discount code is valid for 4 months after issue.

We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even if the antibody turns out to be unsuitable for the species, you will still receive the discount on your next purchase after your Abreview has been submitted.
