  • Function

    After binding acetylcholine, the AChR responds by an extensive change in conformation that affects all subunits and leads to opening of an ion-conducting channel across the plasma membrane.
  • Involvement in disease

    Defects in CHRNB2 are the cause of nocturnal frontal lobe epilepsy type 3 (ENFL3) [MIM:605375]. ENFL3 is an autosomal dominant epilepsy characterized by nocturnal seizures with hyperkinetic automatisms and poorly organized stereotyped movements.

      Immunocytochemistry/ Immunofluorescence - Anti-Nicotinic Acetylcholine Receptor beta 2/CHRNB2 antibody (ab41174)
      ICC/IF image of ab41174 stained SHSY5Y cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab41174, 1/1000 dilution) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

      Additional bands at: 46 kDa. We are unsure as to the identity of these extra bands.


      Exposure time: 3 minutes


      The band observed at 52 kDa could potentially be a cleaved form of Nicotinic Acetylcholine Receptor beta 2/CHRNB2 due to the presence of a 25 amino acid signal peptide.

    Question

    I have been incubating in primary antibody at 4 degrees overnight in TBS-T with 1% BSA. I will try the secondary alone condition, but we have used this secondary antibody successfully with other primary antibodies, so I do not think that is the issue. Also, I will try a longer blocking period and more washes. On the datasheet for this antibody there isn't an image of a representative western blot using Abcam ab41174. Could you please send me an image of the band that Abcam has been able to detect by western blot? Also, is there anything else you can suggest for me to try? Or a positive control that you could send me to work with?

    Thank you for your enquiry. I have added two of the three following articles using this antibody to our datasheet (I have included a datasheet link below for your convenience). The 1997 article is not available via the internet, but is the original source of the antibodies in the literature. In the Nguyen article, there is a nice picture of a western blot. References: 1.
