Application Abreviews Notes
WB 1/5000. Detects a band of approximately 149 kDa (predicted molecular weight: 149 kDa).

  • Function

Play a major role in cholesterol homeostasis. Is critical for the uptake of cholesterol across the plasma membrane of the intestinal enterocyte. Is the direct molecular target of ezetimibe, a drug that inhibits cholesterol absorbtion. Lack of activity leads to multiple lipid transport defects. The protein may have a function in the transport of multiple lipids and their homeostasis, and may play a critical role in regulating lipid metabolism.
  • Tissue specificity

Widely expressed. Expressed in liver. Also expressed in small intestine, pancreas, kidney, lung, pancreas, spleen, heart, gall bladder, brain, testis, stomach and muscle.
  • Sequence similarities

    Belongs to the patched family.
    Contains 1 SSD (sterol-sensing) domain.
  • Post-translational

    Highly glycosylated.
  • Cellular localization

Apical cell membrane. Cell membrane. Cytoplasmic vesicle membrane. Subfractionation of brush border membranes from proximal enterocytes suggests considerable association with the apical membrane fraction. Exists as a predominantly cell surface membrane expressed protein (By similarity). According to PubMed:15671032, localizes in a subcellular vesicular compartment rich in RAB5.

  • Anti-Niemann Pick C1 Like 1/NPC1L1 antibody [EPR5717] (HRP) (ab201773) at 1/5000 dilution + Liver (Human) Tissue Lysate - fetal normal tissue at 10 µg

    Developed using the ECL technique.

    Performed under reducing conditions.

    Predicted band size: 149 kDa
    Observed band size: 149 kDa

    Exposure time: 12 minutes

This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% milk before being incubated with ab201773 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406.
