Target

  • Function

    NMDA receptor subtype of glutamate-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. Mediated by glycine. This protein plays a key role in synaptic plasticity, synaptogenesis, excitotoxicity, memory acquisition and learning. It mediates neuronal functions in glutamate neurotransmission. Is involved in the cell surface targeting of NMDA receptors.
  • Sequence similarities

    Belongs to the glutamate-gated ion channel (TC 1.A.10.1) family. NR1/GRIN1 subfamily.
  • Post-translational
    modifications

    NMDA is probably regulated by C-terminal phosphorylation of an isoform of NR1 by PKC. Dephosphorylated on Ser-897 probably by protein phosphatase 2A (PPP2CB). Its phosphorylated state is influenced by the formation of the NMDAR-PPP2CB complex and the NMDAR channel activity.

    Immunocytochemistry/ Immunofluorescence analysis of mouse primary neuron cells labeling NMDAR1 with purified ab109182 at 1/50 (9.5µg/mL). Cells were fixed in 4% paraformaldehyde and permeabilized with 0.1% Triton X-100. Cells were counterstained with Ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1/200 (2.5 µg/mL). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1/1000 (2 µg/mL) dilution. DAPI (blue) was used as nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.
    Confocal scanning Z step was set as 0.3 µm followed by image processing with maximum Z projection.

That is correct, according to Uniprot the exact immunogen sequence is missing in isoforms 1, 2 and 7 but conserved in isoforms 3 (also known as long, NR-1-3), 4, 5 and 6.

Unfortunately, we do not have the epitope information for this antibody but according to the immunogen sequence, it is towards the cytoplasmic region of the membrane. Therefore, this product may not be suitable for live cells in flow cytometry.

Thank you for your enquiry.

This product was tested with permeabilized SH‐SY5Y cells and the cells were fixed with 2% paraformaldehyde and incubated at room temperature for 10 min. Our labs use the intracellular staining protocol only, and so there is no guarantee if this antibody can be used with live cells.

In general, it should not be a problem but should you not see expected results, please try with fixed cells.

Question

I have found an antibody from another company called "rabbit anti-NR1pan" in the paper : "Newborn- and adult-derived brain microvascular endothelial cells show age-related differences in phenotype and glutamate-evoked protease release" by Legros et al. in Journal of Cerebral Blood Flow & Metabolism (2009) 29, 1146-1158. Unfortuneately, the company stopped selling this antibody.

Thus, I wanted to ask whether you can offer a good primary antibody against the NR1 subunit of the NMDAR (NMDARNR1, Grin1, NMDAR1) for western blotting and immunofluorescence. This antibody has to react with mouse and recognize the isoforms of NR1 that means it should be a PAN-antibody, the preferred species would be rabbit.

We have a few rabbit-raised antibodies to NMDAR1 that were tested in mouse as well as WB and ICC.

ab28669 (https://www.abcam.com/NMDAR1-antibody-ab28669.html), but based on the immunogen sequence, it would only recognize only isoform 1 in mouse.

ab109182 (https://www.abcam.com/NMDAR1-antibody-EPR2481-2-ab109182.html). Here I currently have no detailed information about the location of the immunogen peptide, and will inquire with the lab about that. I will let you know when I hear back from them.

ab17345 (https://www.abcam.com/NMDAR1-antibody-Neuronal-Marker-ab17345.html). This antibody was raised against a peptide in the N-terminal region which seems to be conserved among the isoforms. The antibody has not yet been tested inmouse, but is predicted to react based on sequence homology. In that case I can offer you a testing discount code for trying it in mouse.
