This antibody produced a thin membrane bound staining in a subpopulation of neurones, such as cortical neurons. The picture shows the staining obtained at the level of the cerebral cortex. I am surprised that there are not many more neurons stained. According to the Allen brain, many neurons should be stained, which is not the case.
The sections used came from animals perfused fixed with Paraformaldehyde 4%, in phosphate buffer 0.2M. Following postfixation in the same fixative overnight, the brains were cryoprotected in sucrose 30% overnight. Brains were then cut using a cryostat and the immunostainings were preformed using the ‘free floating’ technique.
