      Question

      How are you?

I thought this may interest Abcam.

Please see below the response from our customer:

I have a proposal for your supplier. I am more than happy to write a review in return for a new supply. We have performed both IHC and WB with this antibody and we have fantastic results. By comparison, the IHC images given in your data sheet is not convincing. It look like H&E staining rather than and IHC staining. If your supplier is interested, I am more than happy for further talks.

I look forward to receiving your response!

      Kind regards,

      Answer

Thank you for forwarding this enquiry.

Reviewing the IHC images from the datasheet, I understood the customers concerns and contacted the testing laboratory. The images were provided tothem by a collaborator who uses red dyes for detection. In the human heart you can see darker pink/red areas of cytoplasmic staining. However, unfortunately it is difficult to tell with the skeletal muscle tissue. They are going to retest the antibody inIHC with standard DAB detection.

I am sorry we are not able to offer anything further for this customer on this occasion as the antibody is already tested and covered by our guarantee in IHC-P. We are happy to reassure our customers that all of our products are covered by our Abpromise, which guarantees that the product will work in the applications and species specified on the datasheet, or we will offer a replacement, credit, or refund within 6 months of purchase.

Otherwise, we like to encourage all of our customers to submit an Abreview via the online product datasheet and the customer may like to consider this in return for Abpoint rewards. We always appreciate customer feedback, whether positive or negative, and we make all product information available to researchers. Each Abreview earns Abpoints that can be used for discounts on future purchases or rewards such as Amazon.com gift certificates.

      Question

      Dear Tech support,

      How are you?

I have a question from a customer and may require your assistance on this please; please see below:

I am planning to use one of your antibodies (ab 2002-100), nucleolar protein 30 (NOP 30). As you know, NOP30 is the product of the NOL3 gene. Due to alternative splicing, NOL3 produces two protein products, ARC and NOP 30. In the literature, NOL3, ARC and NOP30 are considered synonyms. Your datasheet says the same. Given that the C-terminal region of ARC is proline/glutamic acid (P/E) rich and that of NOP30 is serine/arginine (S/R) rich, I am not sure if it is appropriate to consider both as synonyms. My question, therefore, is what will I be detecting? Is it NOP30 or ARC?

      Please advise!

      Kind regards,

      Answer

      Thank you for your enquiry.

Based on the customers question I would define ab2002Anti-Apoptosis repressor with CARD antibodyas specific to ARC rather than NOP30. Looking at the two isoforms on UniProt the antibody is specific to isoform 2 as listed under O60936.

I hope this will be helpful to you. If you have any further questions, please do not hesitate to contact us.
