  • General notes

Normal goat serum ab7481 is used extensively for the blocking of non-specific antibody binding in tissue and cell staining, and in other applications of antibodies.

The goat serum blocks the binding of Fc receptors in the sample to the primary and secondary antibodies used in the experiment, and also blocks non-specific binding of the antibodies to the sample.

Typically the serum used for blocking is from a different species than the species in which the primary antibody was raised. Often the blocking serum is from the species in which the secondary antibody was raised.

Serum can be used directly for blocking, or as a constituent of a blocking buffer.

    Strain: Mixed breed and sex.

    Raised in: Goat

    Purity: Whole antiserum

This product is for research use only and not intended for diagnostic or therapeutic use of any kind.

Our Abpromise guarantee covers the use of ab7481 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

  • Cells grown in 8-well chamber slides to 70% confluence were fixed with 4% paraformaldehyde (15 min at room temperature (RT)), and permeabilized with 100% ice cold methanol (10 min at 20°C). The slides were incubated with 10% normal goat serum (ab7481) in PBS for 1 hr, and incubated with anti-maspin (1:100) antibody alone or in a combination with either anti-lamin B (1:50), anti-HDAC1 (1:50) or anti-GRP78 (1:50) at 4°C overnight. Cells were washed and incubated for 2 hrs at room temperature (RT) with Alexa Fluor 488 (1:500) alone or in combination with Alexa Fluor 594 (1:500). The nuclei were counterstained with DAPI.
DU145 cells infected with adenovirus expressing either maspinWT or maspinD346E Confocal immunofluorescence imaging of maspin (green) and nuclear envelope marker lamin B (red) in DU145 cells after infection.

  • Slides were pretreated with Hydrogen Peroxide Block followed by 15% normal goat serum (Abcam, Cambridge, UK) for 1 hour. The primary antibody, anti-PTX3, N-terminal antibody produced in rabbit was diluted 1:300 in PBS with 2.5% goat serum (ab7481), applied to slides and incubated for 3 hours at 4°C. The primary antibody was omitted in the negative controls. The PTX3 binding was revealed using a universal secondary antibody polymer formulation conjugated to horseradish peroxidase (HRP). The HRP activity was subsequently visualized with diaminobenzidine (DAB) substrate/chromogen and counterstaining with hematoxylin was performed.

There is no need to heat-inactivate serum if you are using it as a blocking reagent before staining fixed cells or tissue. Heat-inactivation is only necessary if you are using the serum for tissue culture. (The heat inactivates complement, and this eliminates complement-mediated cell lysis).

ab138478 is sterilized, whereas ab7481 is not. That is the only difference between the two products and the reason that ab138478 is more expensive.

ab1002 Anti-Rabies Virus antibody looks very promising as it has been established for the use in IHC-P and should detect a wide range of rabies strains. As a starting dilution the customer can try a 5 ug/ml dilution incubated overnight at 4C. However, it may be that the actual optimum dilution may be lower or higher, but this depends on her samples. Another point is that I would strongly recommend a heat induced Antigen Retrieval if her slides are fixed with formalin.

https://www.abcam.com/index.html?datasheet=1002 (or use the following: https://www.abcam.com/index.html?datasheet=1002).

A matching secondary antibody would be Goat polyclonal Secondary Antibody to Mouse IgG2a - heavy chain (Biotin), pre-adsorbed (ab98696)

https://www.abcam.com/index.html?datasheet=98696 (or use the following: https://www.abcam.com/index.html?datasheet=98696).

To be fair, I am not the biggest fan of ready- to- use solution (andmost of my colleagues here a neither), for the following reason: most of all antibodies need optimization, may it be the blocking step, the retrieval time or method, the antibody incubation buffer etc. With ready-mix solutions it almost impossible to adjust the experiment settings. Ready mix solution make sense if the antibody is used in a high -throughput lab (like 20 slides at once with a fixed protocol), but not if you have to "learn" IHC-P and have the freedom to adjust the protocol fore the perfect results. In addition, it's cheaper ;)

Of course, once the protocol is optimized, using ready mixed solution are a very time efficient and make life easier, but I wouldn't use them as a beginner.

All the buffers your customer needs (see attachment) are listed in our very detailed and easy to follow step-by-step our protocol. However, none of my advice is written in stone, so if your customer prefers the ready mix solutions, we are more than happy to sell them to her.

Something we cannot provide are the reagents for the de-paraffinization, I am afraid, but a company like Sigma should be able to help with that.

Pease sent the customer our protocol and my advice. I am more than happy to assist her to get her experiments running. And another thing: she shouldn't be shy to ask any question she may have about the protocol!

I hope this more information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

It is my pleasure to help.

Thank you for contacting us.

We presently have only 1 product against mouse aggrecan, ab36861. This is a rabbit polyclonal. In ICC and appropriate secondary would be anti-Rabbit IgG with a fluorescent tag which could be excited by your lasers. A product such as ab96899,Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (DyLight® 488),https://www.abcam.com/Goat-polyclonal-Secondary-Antibody-to-Rabbit-IgG-H-L-DyLight-488-pre-adsorbed-ab96899.html

We currently have 20 products against collagen II. However taking into account that this will have to be used in mouse tissue in conjunction with an anti-aggrecan rabbit polyclonal we can limit this to ab3092Anti-Collagen II antibody [5B2.5]. This is a mouse monoclonal antibody of isotype IgG2a that has been tested inIHC-Fr,IHC-P,WB and IF. An appropriate secondary should preferably be a F(ab) fragment as mouse on mouse staining can be difficult, please see this link for more information about mouse on mouse staining,https://www.abcam.com/index.html?pageconfig=resource&rid=11465. One such product would be ourGoat F(ab')2 polyclonal Secondary Antibody to Mouse IgG - (Fab')2 (DyLight® 550), pre-adsorbed,https://www.abcam.com/Goat-F-ab-2-polyclonal-Secondary-Antibody-to-Mouse-IgG-Fab-2-DyLight-550-pre-adsorbed-ab98758.html

Abcam also offers a number of ICC and IHC reagents. As both of the secondary antibodies listed above are raised in goat, we recommend using a 5-10% Goat serum as your blocking reagent and antibody incubation solution, ab7481https://www.abcam.com/Goat-Serum-ab7481.html. We, also offer buffers and antigen retrieval solutions,https://www.abcam.com/index.html?pageconfig=resource&rid=12854, as well as mounting media for fluorescent staining ab103748,https://www.abcam.com/BrightMount-Plus-25ml-Aqueous-Mounting-Medium-Anti-fading-for-Fluorescent-Staining-ab103748.html.
