      Immunohistochemical analysis of formalin-fixed, paraffin-embedded Human ovarian endometriosis tissue, staining NQO1 with ab97385.

      Antigen retrieval was performed in sodium citrate buffer in a microwave oven. Samples were blocked with rabbit serum for one hour before incubating overnight with primary antibody (1/200). Stainined was detected using DAB.

    Question

    I’ve tested the antibody NQO1 (ab97385) which you send as a replacement for the previous one and I’m still not satisfied with the results. I attached a full report on my experiments so that you can revise it carefully and let me know if I did something wrong. I detected NQO1 together with additional bands in human tissues as well as in two cell lines: HepG2 and JEG3. In cell lines, NQO1 seems to be present in 2 isoforms, while in tissue an additional bend size 50 kDa was detected as well. We found no reports about the existence of different NQO1 isoforms; however, we detected additional bands in all tissue samples examined. HepG2 is often used as a control for NQO1 and even there, 2 bands were seen. Do you have any new information about NQO1 or is this antibody simply not specific enough. Please give me some further suggestions or ideas on how to optimize my experiments to get reliable results.

    Reply from the lab and the image is attached: "Our QC image also shows there are double bands in HepG2 cell line, and the pattern of these two bands are similar with the double bands with ~31KDa and ~26KDa detected in HepG2 by the customer. We believe the size difference of double bands between our QC image and the customer's may result from the inaccuracy of size labeling or different brand MW marker. Based on HPRD, NQO1 has 3 isoforms(predicted size : 30.87KDa, 27.3KDa, and 26.37KDa respectively ) and 3 Acetylation site. http://www.hprd.org/summary?hprd_id=00518&isoform_id=00518_1&isoform_name=Isoform_1 And GTX100235 may cross with all 3 isoforms based on immunogen blast data. Therefore, the double band in HepG2 may be different isforms detected by GTX100235 NQO1 antibody. Besides, we have not found any ~50KDa in all cell lines we have tested."
