IHC-FoFr 1/200. PubMed: 113319561/200. Tissue was immersion fixed (not perfusion fixed) overnight in 4% paraformaldehyde/PBS pH 7.4 and then cryoprotected by incubation in 30% sucrose overnight. Sections were blocked for 1 hour with 1% goat serum/5% BSA in PBS with 0.02% sodium azide and 0.02% Triton X-100 (see PubMed: 11331956).

    Cell adhesion, ankyrin-binding protein involved in neuron-neuron adhesion. May play a role in the molecular assembly of the nodes of Ranvier.

Yes, Karen and I spoke and decided on the advice Karen gave to you. I have still have the same comments. What results did you obtain from the no primary control you ran? If you are still seeing the high background with the secondary only, have you tried a different secondary? The SuperSignal detection system is extremely sensitive, and it may be necessary to dilute out the primary antibody further than you have. Have you tried using ECL+ or ECL instead of SuperSignal? In addition, we would let our blots stand before exposing to film since the signal was so strong. I would suggest either trying a less sensitive ECL system, using less primary antibody (try 1/5000), or letting the blot stand for a while before exposing them to the film. I am not surprised that other antibodies have worked well in this system because super signal is a good system. I am concerned that the antibody was optimized at 1/500 dilution + 20 µg of postnatal day 8 mouse brain extract in SDS using ECL plus and under the conditions you have tried with the supersignal system you have a completely black blot suggesting overexposure, too much antibody, too much supersignal etc. I really believe you need to optimize the antibody based on our suggestions.

I would like to suggest the following modifications to your protocol: Please use a fresh blot and try: blocking in 5% milk for 30 min, incubating in primary overnight in 5% milk at 4 degrees, and then incubate in secondary for 1 hour at room temperature.
