WB Use a concentration of 1.25 µg/ml. Detects a band of approximately 26 kDa (predicted molecular weight: 26 kDa). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 4 - 8 µg/ml.

  • Function

    Component of the cleavage factor Im (CFIm) complex that plays a key role in pre-mRNA 3'-processing. Involved in association with CPSF6 or CPSF7 in pre-MRNA 3'-end poly(A) site cleavage and poly(A) addition. NUDT21/CPSF5 binds to cleavage and polyadenylation RNA substrates. The homodimer mediates simultaneous sequence-specific recognition of two 5'-UGUA-3' elements within the pre-mRNA. Binds to, but does not hydrolyze mono- and di-adenosine nucleotides. May have a role in mRNA export.

I have read the details, you have kindly provided and have few suggestions; these might improve the results.

- NUDT21 as per http://www.genecards.org/cgi-bin/carddisp.pl?gene=NUDT21exist as a component of the cleavage factor Im (CFIm) complex, composed of, at least, NUDT21/CPSF5 and CPSF6 or CPSF7. Within the cleavage factor Im complex, the NUDT21/CPSF5 homodimer is at the core of a heterotetramer. It is likely that the band at 55 kDa is a homodimeric form of NUDT21 however if the protein has been reduced without a doubt then the possibility of this being a homodimer can be ruled out. Could you however try heating the lysates at 100C for 10 minutes and carefully checking the reducing agents used?

- The NUDT21 protein is also heavily glycosylated so the observed band size (perhaps faint 40kDa band) would be different than predicted. (http://www.uniprot.org/uniprot/O43809). We have observed 26kDa band size in A-431 cell line though so this cell line lysates might be used as positive control.
