    To whom it may concern:

    I am troubleshooting using your monoclonal antibody ab109180 to identify Nur77 expression in human PBMCs. In mouse, Nur77 has been identified as a marker analogous to CD69, induced upon TCR signaling. I have tried using the antibody in a time course experiment in both flow cytometry and western blots. I was wondering if you can offer me a positive control or a peptide to block antibody binding so I can ask the question, does Nur77 serve as a read out of antigen stimulation in human T cells?

    Answer

    Thank you for your email.
    Unfortunately we are not able to provide the immunogen peptide for this particular antibody, as the laboratory providing this antibody does not have it available. I am very sorry.
    In regards to positive control, I can recommend HepG2 cells, as they have provided good staining as shown on the datasheet. Otherwise, for mouse leukocytes, PMA and ionomycin treatment can induce Nur77 expression. I am not sure whether a similar stimulation could also induce expresssion on your cells?
