  • Function

    Mediates the Na(+)-independent uptake of organic anions such as pravastatin, taurocholate, methotrexate, dehydroepiandrosterone sulfate, 17-beta-glucuronosyl estradiol, estrone sulfate, prostaglandin E2, thromboxane B2, leukotriene C3, leukotriene E4, thyroxine and triiodothyronine. May play an important role in the clearance of bile acids and organic anions from the liver.

    Highly expressed in liver. Not detected in heart, brain, placenta, lung, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis, ovary, small intestine, colon and leukocyte.

I am glad that the customer has received the replacement product. I can confirm that this antibody has detected a band of ˜85 kDa in liver basolateral membrane as well as a band of ˜90 kDa in transfected HEK293 cells. Some difference in the band size can probably be expected due to differential post-translational modifications. The protein treated toremove theglycosylation(usingthe enzymeN-glycosidaseF) has been reported to be detected at around 60 kDa.

The protocol used previously should be fine. Some optimisation may be required in order to achieve the optimum signal to background. Please do bare in mind that the antibody is of the isotype IgM and the appropriate secondary antibody needs to be used. Please do let me know how they geton.

Thank you for your enquiry. Both antibodies were tested in IP using human liver samples (~20-50 ug) and about 1 ug/ml of antibody used. For IF, human liver and transfected HEK293 whole cell lysates were used. Ab15442 was used at a dilution of 1:20. The fixative was 4% PFA for ~10-15 minutes. Ab15441 was used at 1:100 in IF.
