  • Relevance

    Ochratoxin A is a polyketide-derived dihydroiso-coumarin moiety linked through the 12-carboxy group to phenylalanine. Ochratoxin A is the most important and most commonly occurring of a structurally related group of compounds. It is produced by some species of Aspergillus, such as A. ochraceus, mainly in tropical regions, and by Penicillium verrucosum, a common storage fungus in temperate areas such as Canada, eastern and north western Europe and parts of South America. Ochratoxin A is a potent toxin affecting mainly the kidneys, in which it can cause both acute and chronic lesions, whereas its dechloro derivative, ochratoxin B, is non-toxic. A nephrotoxic effect has been demonstrated in all mammalian species.

The antibody ab23965 has been tested on Ochratoxin A from Sigma, Ref. O 1877. http://www.sigmaaldrich.com/catalog/search/ProductDetail/SIGMA/O1877 The following protocol was used: Ochratoxin A is coupled on BSA and used as antigen. -This antigen is coated in a 96 wells plate (EIA/RIA high binding Ref. Corning Costar 3590) at ~ 10µg/ml in bicabonate buffer pH 9.5. Incubation 1 hour at 37°C with shaking. - Saturation : coupling solution is eliminated by turning over the plate. Add 150 µl of BSA 0.5% in PBS. Incubation 1/2 hour at 37°C with shaking. - Eliminate the BSA solution. Add 0.1ml of ab23965 diluted at 1/5000 in PBS/Tween 20(0.1 %). Incubation 1/2 hour to 1 hour at 37°C with shaking. - Wash the plate 4 time with PBS/Tween 20 (0.1%) or TBS/Tween 20 (0.1%). Add 0.1ml of diluted secondary antibody. Incubation 1/2 hour at 37°C with shaking. - Wash the plate 4 time with PBS/Tween 20 (0.1%) or TBS/Tween 20 (0.1%). Add 0.1ml Chromogen. (to be chosen according the enzyme used for labelling the secondary antibody). -Stop the reaction. -Read the OD of the wells.
