    Flow Cyt: 1/50.
    ICC: 1/250 - 1/500.
    IHC-P: 1/250 - 1/500. Antigen retrieval is recommended.
    WB: 1/1000 - 1/5000. Detects a band of approximately 58 kDa (predicted molecular weight: 51 kDa).

    • Function

      Transcription factor that specifically binds to the octamer motif (5'-ATTTGCAT-3'). Regulates transcription in a number of tissues in addition to activating immunoglobulin gene expression. Modulates transcription transactivation by NR3C1, AR and PGR. Isoform 5 activates the U2 small nuclear RNA (snRNA) promoter.
    • Tissue specificity

      Isoform 3 is B-cell specific. Isoform 5 is expressed in B-cells and the immunoglobulin-expressing T-cell line MOLT-4, but not in the T-cell line BW5147.

      why is the actual band size different from the predicted?

    • ab92458 at a 1:250 dilution staining Oct-2 in human tonsil (paraffin embedded section) by immunohistochemistry.

      Perform heat mediated antigen retrieval with EDTA buffer pH 9 before commencing with IHC staining protocol.

    • Overlay histogram showing Raji (Human Burkitt's lymphoma cell line) cells stained with ab92458 (red line).

      The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab92458, 1:50) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG (H+L) (ab96899) at 1:500 dilution for 30 min at 22ºC.

      Isotype control antibody (black line) was rabbit IgG (monoclonal) (1 µg/1x106 cells) used under the same conditions.

      Acquisition of >5,000 events was performed.
