The stock solution of the Optiblot DTT Reducer (10X) ab119199 in 2M. This means that once diluted 10 times this produces a 200 mM solution. Therefore if you usually use 10 mM with your samples I would suggest diluting this 200 times. The higher concentration should not really have an effect of the running of protein gels but as the pKa is around 8, in high concentration it can alter the pH somewhat which may affect the migration. We would normally suggest using 50 mM to ensure fully reducing the disulphide bonds in your samples.

Answer

I now have further information in regards to the storage of the Optiblot DTT Reducer (ab119199). I can confirm that this should be fine andshould remain stable for longer if stored at -20°C. I would recommend dividing the 1 mL into smaller aliquots and freezing at -20°C until needed.

I have not seen that we have tested or have any additional data regarding the dimerization of this protein or which reducing agents might be more effective. We generally use a 2x Laemmli buffer with 4% SDS, or use our Optiblot systems when testing. I have place both the recipe for the standard 2x Laemmli buffer and links to our Optiblot page and a few recommended products below.

2x Laemmli:

4% SDS

10% 2-mercaptoethanol

20% glycerol

0.004% bromophenol blue

0.125 M Tris-HCl

Check the pH and adjust pH to 6.8.
