  • ab87312 staining oxytocin receptor in MCF7 cells treated with oxytocin (ab120186), by ICC/IF. Increase of cytoplasmatic oxytocin receptor localization and correspondent decrease in nuclear oxytocin receptor correlates with increased concentration of oxytocin, as described in literature.
    The cells were incubated at 37°C for 15 minutes in media containing different concentrations of ab120186 (oxytocin) in DMSO, fixed with 4% formaldehyde for 10 minutes at room temperature and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with ab87312 (5 µg/ml) was performed overnight at 4°C in PBS containing 1% BSA and 0.1% tween. A DyLight 488 donkey anti-goat polyclonal antibody (ab96931) at 1/250 dilution was used as the secondary antibody. Nuclei were counterstained with DAPI and are shown in blue.

As we do not currently undertake our own research I can only offer advice regarding the literature. The main difference between the free acid form and the non-free acid is a difference to the terminal end of the peptide. The non-free acid has a -NH2 group at its terminal end whereas Oxytocin (free acid) has an –OH group. This is likely to affect the biological activity but looking at published research, Oxytocin (non-free acid) is much more commonly published on. Sorry I could not provide more specific information regarding the free acid.

We have not specifically tested ab52147 on C2C12 cells. However, we do have customer feedback regarding the use of this antibody to detect the phosphorylated protein in treated myometrial smooth muscle cells.

In his treatment, this customer used oxytocin, which we carry as ab120186 and phorbol 12-myristate 13-acetate (PMA), which we carry as ab120297.

I do not know enough about the protein to tell you definitively whether or not this protein is endogenously phosphorylated in C2C12 cells. I do have an image of ab52174 used in WB with untreated RAW264.7 cells though, which we sell as ab7187. I'll be sure to get that WB image added to the datasheet online, but it will take a few days to appear.
