  • Function
    Catalyzes the citrullination/deimination of arginine residues of proteins. Citrullinates histone H3 at 'Arg-8' and/or 'Arg-17' and histone H4 at 'Arg-3', which prevents their methylation by CARM1 and HRMT1L2/PRMT1 and represses transcription. Citrullinates EP300/P300 at 'Arg-2142', which favors its interaction with NCOA2/GRIP1.
  • Tissue specificity
    Expressed in eosinophils and neutrophils, not expressed in peripheral monocytes or lymphocytes.
  • Involvement in disease
    Genetic variations in PADI4 are a cause of susceptibility to rheumatoid arthritis (RA) [MIM:180300]. It is a systemic inflammatory disease with autoimmune features and a complex genetic component. It primarily affects the joints and is characterized by inflammatory changes in the synovial membranes and articular structures, widespread fibrinoid degeneration of the collagen fibers in mesenchymal tissues, and by atrophy and rarefaction of bony structures. Note=Could have an important role in the pathogenesis of rheumatoid arthritis by increasing citrullination of proteins in rheumatoid arthritis synovial tissues, leading, in a cytokine-rich milieu, to a break in tolerance to citrullinated peptides processed and presented in the appropriate HLA context.

  • ICC/IF image of ab50247 stained HeLa cells. The cells were 4% formaldehyde (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab50247, 1/200 dilution) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

The PAD4 protein is predicted to run around 74 kDa based on the molecular weight of the amino acid sequence, however full length PAD4 has been shown to migrate around 67 kDa in SDS-PAGE:


The exact cause of this has not been determined, but some proteins do not run at their predicted molecular weights due to structure and/or amino acid charge. Looking through our other antibodies to PAD4, they do recognize a band below 70 kDa. Based on this information, the band on your blot around 67 kDa is most likely the specific PAD4 band.

The exact concentration is necessary when you would like to use Isotype control in IHC or Immunoprecipitation other than that it is OK.

Please note that we can only provide exact concentration for ab128089 because it is purified antibody. We cannot provide concentration for ab50247 because it is a un-purified antibody.

There are a few variations that may work:

1 - Depending on the blocking agent you are using, you could try using 5% BSA, as this may help reduce the background levels that you are seeing.

2 - If you are seeing the band of interest, but also getting high background, then using a lower concentration of the antibody may be helpful in reducing background..

3 - Another, way to decrease background is to use ab126587, our https://www.abcam.com/10X-Blocking-Buffer-ab126587.html, but I would only recommend that if using the BSA does not prove to be successful.
