  • Function
    PTH elevates calcium level by dissolving the salts in bone and preventing their renal excretion. Stimulates [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblastic cells.
  • Involvement in disease
    Defects in PTH are a cause of familial isolated hypoparathyroidism (FIH) [MIM:146200]; also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterized by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. FIH exist both as autosomal dominant and recessive forms of hypoparathyroidism.

  • ab14498 staining Parathyroid Hormone in canine parathyroid tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue was fixed with formaldehyde and a heat mediated antigen retrieval step was performed using the microwave method. Samples were then blocked with 10% serum for 20 minutes at room temperature followed by incubation with the primary antibody at a 1/100 dilution for 50 minutes. An undiluted biotin-conjugated goat anti-mouse polyclonal was used as secondary antibody.

I'm not sure how the IgG concentration can be specifically measured with a spectrophotometer if there were other proteins in the vial. All proteins (containing aromatic residues) will absorb at 280nm. The only method I'm aware of for measuring IgG concentration in a solution with other proteins (such as serum or tissue culture supernatant) would be by ELISAagainst an IgG standard curve. I've discussed this with my colleagues, and we're wondering if the user has a reference for this method, or could he/she describe the protocol?

The antibody has been protein G purified and no proteins are added, so we expect it to be quite pure andintend to provide100 ug of IgG per vial. I see that the 3 new vials were delivered today, so if theend user does wish to test these vials then please keep me updated of the results. If the measurements are also coming up short or if the user prefers not to test them in light of the QC information, please let me know and we can arrange a credit or refund.

I've heard back from the lab, and each new lot of antibody is tested after production (i.e. before lyophilization) using a Bradford assay and then is tested again after lyophilizaiton.
